Graft-versus-host disease (GvHD) is a common and potentially serious complication that occurs after allogeneic hematopoietic stem cell transplantation (HSCT).
HSCT is used as a potential cure for hematological cancers, syndromes of bone marrow failure, and congenital disorders of the hematopoietic system (Hymes, Alousi and Cowen, 2012; Passweg et al., 2025). GvHD occurs when the donor stem cells or the graft attack healthy cells in the patient or the host (Blood Cancer United, 2024). GvHD can damage a patient’s tissues and organs, skin, eyes, mouth, hair, nails, liver, esophagus, intestines, joints, lungs, and genitals (Blood Cancer United, 2024). There are two main types of GvHD: acute GVHD (aGvHD), and chronic GvHD (cGvHD).
In the 7MM, the number of diagnosed incident cases of first allogeneic HSCT is expected to increase by 13.91% over the forecast period at an annual growth rate (AGR) of 1.39% from 23,192 cases in 2025 to 26,418 cases in 2035. In 2035, the US will have the highest number of diagnosed incident cases of first allogenic HSCT in the 7MM with 11,087 cases, and France will have the fewest diagnosed incident cases of first allogenic HSCT with 1,590 cases. In the 7MM, the number of three-year diagnosed prevalent cases of aGvHD is expected to increase by 15.87% over the forecast period at an AGR of 1.59%, from 21,968 cases in 2025 to 25,454 cases by 2035. In 2035, the US will have the highest number of three-year diagnosed prevalent cases of aGvHD in the 7MM with 11,670 cases, and France will have the fewest three-year diagnosed prevalent cases of aGvHD with 1,494 cases. In the 7MM, the number of three-year diagnosed prevalent cases of cGvHD is expected to increase by 15.91% over the forecast period at an AGR of 1.59%, from 25,768 cases in 2025 to 29,867 cases by 2035. In 2035, the US will have the highest number of three-year diagnosed prevalent cases of cGvHD in the 7MM with 13,824 cases, and France will have the fewest three-year diagnosed prevalent cases of cGvHD with 1,769 cases. GlobalData epidemiologists attribute the increase in diagnosed incident and three-year diagnosed prevalent cases of aGvHD and cGvHD partly to the moderately rising trend of increase in transplantation in the 7MM.
Scope
This report provides an overview of the risk factors, comorbidities, and the global and historical epidemiological trends for GvHD in the seven major markets (7MM: US, France, Germany, Italy, Spain, UK, and Japan). The report includes a 10-year epidemiology forecast for the diagnosed incident cases of first allogeneic HSCT, diagnosed incident cases of aGvHD, and diagnosed incident cases of cGvHD. The diagnosed incident cases of aGvHD and cGvHD are segmented by overall age (0–18 years and above 18 years), donor types, and three-year diagnosed prevalent cases. The diagnosed incident cases of aGvHD are further segmented by Glucksberg severity grades, and 100-day mortality. The diagnosed incident cases of cGvHD are further segmented by severity based on National Institutes of Health (NIH), and one-year mortality. This report focuses on aGvHD segmented by severity, defined by the Mount Sinai Acute GvHD International Consortium (MAGIC), and cGvHD segmented by severity by NIH. This definition is applied consistently across all markets. This epidemiology forecast for GvHD is supported by data obtained from peer-reviewed articles and population-based studies. The forecast methodology was kept consistent across the 7MM to allow for a meaningful comparison of the forecast diagnosed incident cases of first allogeneic HSCT, aGvHD, and cGvHD across these markets.
