PPD is typically managed with psychotherapy (CBT or interpersonal therapy) and antidepressants, most commonly SSRIs, tailored to breastfeeding considerations and patient preferences, alongside practical supports such as sleep and family assistance. For moderate-to-severe illness, faster-acting neuroactive steroid treatments include IV brexanolone (a 60-hour monitored infusion) and oral zuranolone (a short-course regimen), although uptake may be constrained by cost, access, sedation, and logistical burden.
Patients with severe or high-risk features (suicidality, psychosis, or treatment-resistant symptoms) require urgent psychiatric evaluation and may need hospitalization and/or ECT.
The PPD pipeline includes 15 molecules, with no assets in the pre-registration stage, two in Phase III development, and seven in Phase II development.
Over the past decade, 159 clinical trials have been conducted in PPD. The year with the most studies initiated was 2024, with 24 trials, followed by 2021 with 23 trials. Additionally, 2022 saw the most Phase III trials initiated, with five.
Over the past decade, in deals involving companies developing PPD products, acquisitions accounted for the largest share of deals in North America (44.4%). By comparison, licensing agreements were most prevalent in Europe (40%) and partnerships in the Asia-Pacific region (50%). South America, along with the Middle East and Africa, had only asset transactions (100%).
GlobalData survey results indicate SSRIs remain the preferred first-line therapy due to clinician familiarity and perceived effectiveness, while Zurzuvae (zuranolone) is generally positioned as a second-line option for SSRI non-responders and for patients requiring a rapid onset of benefit.
